Research Overview

  • My research program focuses on understanding mechanisms in lung injury, repair, and development. My lab is particularly interested in investigating lung injury caused by toxic chemicals like sulfur mustard and chlorine, acute effects of which can lead to acute respiratory distress syndrome (ARDS). We are investigating pathways that are involved in the pathogenesis of ARDS and are also testing therapies that can mitigate such injuries. Sulfur mustard, a vesicating agent, is increasingly being recognized as a threat agent with exposures resulting in severe morbidity and mortality. Part of this injury results from activation of the coagulation cascade and leaky airways resulting in the formation of bronchial casts. Using aerosolized half mustard to mimic sulfur mustard exposures, we have identified extracellular nucleic acids as major players in causing severe morbidity and mortality. We have also demonstrated that nucleic acid-neutralizing agents can rescue rats from mortality and prevent the progression to ARDS. We have also found that nucleic acid-neutralizing agents are effective in mitigating injury in other models of ARDS.
    Lung injury can be caused by direct insult or indirectly through systemic effects. We are also investigating lung injury caused by arsenicals, another group of vesicants. We have found that arsenicals, caused lung injury following cutaneous exposure to Lewisite. We are also investigating the effect of arsenic trioxide (ATO), a common environmental pollutant and a chemotherapeutic agent, in causing lung injury. We have found that a single dose of inhaled ATO causes acute lung injury.
