Insulin: an allosteric modifier of the sugar carrier mechanism

Abstract

  • Glucose uptake was determined by rat soleus muscle in vitro in Krebs Henseleit bicarbonate buffer containing from 10-28 mM glucose in the presence and absence of insulin (75-5000 μ U/ml). The results indicated that irrespective of the insulin concentration, the effect of insulin was to lower the apparent Km without affecting the V(max). A model is proposed which explains the effect of insulin by postulating the binding of insulin to an allosteric site on the carrier molecule.
